The veteran Serbian player defeated Alcaraz, 16 years his junior, in a tense match that lasted three hours and 37 minutes at the Rod Laver Arena.
The final score stood at 4-6, 6-4, 6-3, 6-4 to Djokovic.
Despite losing the first set, Djokovic managed to rebound after taking a medical time-out to address a left thigh issue.
Following the break, he regained momentum and secured the following three sets against his opponent.
At the conclusion of the match, Djokovic showed gratitude towards his coach Andy Murray, expressing his appreciation for the high-stakes nature of the game.
If successful in the tournament, Djokovic could win an 11th Australian Open title, setting a new record for the most completed by any singles player.
The 37-year-old currently shares the record of 24 major titles with Margaret Court.
In the upcoming semi-finals, Djokovic will meet German player Alexander Zverev, who is targeting his first-ever Grand Slam title.
A win against Zverev would mark Djokovic's 100th victory at the Australian Open, propelling him towards a possible final against world No.1 Jannik Sinner.
Carlos Alcaraz, currently ranked world No.3, had aspirations of becoming the youngest player with a career Grand Slam, having previously won the French Open, Wimbledon, and the US Open.
Alcaraz matched his previous best performance at the Australian Open, having also reached the quarter-finals last year.
